External and Histological Characteristics of Organogenesis from Gladiolus Callus



Abstract
Organogenesis from the gladiolus callus (cv. Topaz and Firebrand) could be made by regulating concentration exogenous auxin and eytokinin, in spite of some difference in accordance with medium pH and cultivars. The rate of shoot and root formation was 100% on the MS medium without growth regulators. The days required for root and shoot formation were 7 and 10 days, respectively, on the MS medium without growth regulators, and this required shorter time than under medium with kinetin. As the result of histological observation, shoots and roots were exogenous and endogenous occurrence, respectively. The shoots, however, were not connected with the roots by a vascular system in a callus. Plantlet of two types was observed, and cormlets were formed at the basal parts of shoot.
